Abstract:

Introduction & Objective: Oral squamous cell carcinoma is the most prevalent malignancy of the mouth which includes more than 90% of oral cancers. LAMP3 is a member of LAMP family and dendritic cell marker located on its membrane. Recent studies showed high expression of this gene in esophageal SCC, head and neck SCC and also cancer of breast, digestive tract and cervix. The objective of this study was to evaluate the expression of LAMP3 gene in oral squamous cell carcinoma and dysplastic oral epithelium by Real-time reverse transcription polymerase (RT-PCR) and also comparing of LAMP3 expression between grade I, II, III OSCC with each other and with dysplastic epithelium.
Method: In this study, 60 samples of squamous cell carcinoma and dysplastic epithelium of oral cavity obtained from the archive of oral and maxillofacial Pathology Department of Mashhad University of Medical Sciences together with their corresponding age and gender information. At the beginning total RNA from all the specimens obtained and gene expression evaluated by Real-time PCR.
Results: In this study 60 sample from 60 patients including 33 female (55%) 27 male (45%) with mean age of 57.7 year and extent of 23-84 years old were evaluated quantitatively for LAMP3 gene expression for grades I, II, III OSCC and dysplastic epithelium result showed that there have been significant difference between different grades of OSCC and dysplastic epithelium. The mean expression of LAMP3 between each grade of OSCC show that grade III had high expression in comparison with grade I, II and dysplastic epithelium and it was almost two times that of grade I and II and also mean expression was lowest in dysplastic epithelium comparing with the OSCC, but there have not been significant difference between grade I and II. The average expression of LAMP3 was higher in OSCC than dysplastic group (p<0.001).
Conclusion: According to our results, higher expression of LAMP3 from dysplastic epithelium to OSCC indicates the process of carcinogenesis in oral mucosa. In higher grades of OSCC with lower differentiation, LAMP3 expression is increasing, so that LAMP3 mRNA in OSCC grade III was higher than OSCC grade I and II.

Abstract:

Dental implants rehabilitations are the most  prefered method of tooth replacement in modern dentistry. Vertical and horizontal  bone deficiencies are the major obstacles of this treatment. Surgical principles of  vertical  and bone grafting, such as stabilization of grafts, space maintenance, and proper wound closure are fort coming points for success.  But determining the most appropriate surgical method for the augmentation of horizontal or vertical insufficiency is the key stone of  the treatment success.
